Description

Context: We are currently seeking a talented and motivated Engineer in Bioinformatics for an exciting project aimed at advancing therapies for sickle cell disease. The candidate will join the lab headed by Drs. S. Azouzi and B. Koehl (Laboratory of Blood group antigens, hematopoiesis, sickle cell disease), at Inserm UMR S1134 at Necker hospital.

 

Background: 

Sickle Cell Disease (SCD) is one of the most common monogenic disorders worldwide. SCD is due to a mutation in the b-globin (HBB) gene, leading to the polymerization of hemoglobin upon deoxygenation and sickling of the red blood cell (RBC). SCD is a multisystem disease characterized by chronic anemia and inflammation leading to painful vaso-occlusive crises, stroke, acute chest syndrome and organ failure. The current laboratory’s project aims to understand the hematopoiesis dysfunction in this disease. The project involves functional and deeper characterization of hematopoietic cells from these patients including HSPCs and mature cells from peripherical blood and blood cord and spleen. Multi-omics data have been obtained including, proteomic, RNAseq, scRNA, scATAC-seq and WES.

 

Mission Objectives: The recruited engineer will be in charge of analysis and integration of "omics" data, including WGS, RNA-Seq, and proteomics from patient cells. Key tasks include analyzing scRNASeq and scATAC-seq data to assess the impact of inflammation, identify target cell populations, and evaluate single-cell responses and immune cell involvement.

 

Required Expertise:

  • Bioinformatic analyses of NGS data (WES, ATAC-seq, RNAseq)
  • Familiarity with scRNA-seq and RNAseq data analysis
  • Statistical analyses of Omics data (GWAS, enrichment analysis, differential gene expression analysis)
  • General knowledge and interest in genetics and molecular biology

Operational Skills:

  • Mastery of Unix/Linux environment and programming languages (Python, R)
  • Proficiency in genomic analysis
  • Knowledge of public genomic databases (NCBI, GenCode, gnomAD, Biomart, etc.)
  • Data management skills for large-scale datasets;
  • Report writing and presentation skills in English and French
  • Enjoy working in a team and collaborating with other scientists, including bioinformaticians.

Desired Experience: 2 to 5 years of research experience

Desired Training: Engineering degree or PhD
